    Screening of the ability of lactic acid bacteria to lower cholesterol in yak yoghurt on Qinghai-Tibet Plateau

    Objective:In this study,202 yak yoghurt samples collected from Tianzhu,Naqu,Zoige,Yushu,Hongyuan and Maqu were screened for excellent strains with high cholesterol-lowering properties.Methods:MRS,M17 and SL medium were selected(MRS medium is the basic medium for culturing lactic acid bacteria,M17 medium and SL medium are the medium for separating lactic acid bacteria,and SL medium is the medium for separating eosinophilic lactic acid bacteria)The separation and purification are carried out,and the catalase test is a negative bacterium.According to the phthalic acid method,the cholesterol is determined,the strain,with the ability to degrade cholesterol is screened,and the 16 S rDNA molecular biology method is used to identify the species.Results:After isolation and purification,a total of 720 strains of lactic acid bacteria were screened.All strains had certain cholesterol degradation ability.There were 52 strains with statistical degradation rate above68%,and the cholesterol degradation rate of one strain of BX39 was as high as 90.05%.After repeated screening,the cholesterol degradation rate of strain BT18 was as high as 96.26%.After 16 S rDNA molecular biology analysis,72 strains of lactic acid bacteria,were found,including64 strains of Lactobacillus reuteri,4 strains of Pediococcus acidilactici,3 strains of Lactobacillus helveticus and 1 strain of Lactobacillus plantarum.Conclusion:Lactobacillus reuteri BT18 has high cholesterol-lowering properties for further research and development.
